What PT-141 (Bremelanotide) is
PT-141, known generically as bremelanotide, is a synthetic cyclic heptapeptide that acts as a melanocortin receptor agonist. Structurally it is a lactam-bridged cyclic analog of alpha-melanocyte-stimulating hormone (alpha-MSH): the ring is closed not by a disulfide but by a lactam (amide) bridge between side chains, which constrains the peptide's conformation. Its closest relative on the research market is Melanotan II (MT-II), the parent compound from which bremelanotide derives — PT-141 was identified as a metabolite of MT-II, and the two molecules differ only at the C-terminus (a free carboxylic acid in PT-141 versus an amide in MT-II). That near-identity, combined with the fact that vendors frequently cross-sell both compounds, makes the PT-141-versus-MT-II question the central identity issue for this peptide. It is sold on the research market without regulated manufacturing or identity guarantees.
Why get your PT-141 (Bremelanotide) independently tested
- PT-141 vs. Melanotan II substitution: the two compounds are routinely sold side by side, share most of their structure, and are easily confused or swapped at the labeling stage. The C-terminal acid-versus-amide difference changes the molecule's polarity and shifts its reversed-phase retention time, so an identity match against a certified bremelanotide reference standard shows which of the two is actually in the vial.
- Cyclization fidelity: the lactam bridge is formed in a dedicated cyclization step during synthesis, and failed or incomplete cyclization leaves linear peptide in the product. The linear analog is a different molecule with different retention behavior, so it registers against both the identity comparison and the purity figure.
- Stereochemistry-related byproducts: bremelanotide's sequence includes a D-phenylalanine residue, and epimerization during synthesis produces diastereomeric impurities that can elute close to the main peak. Chromatographic purity as area percent quantifies how much of the UV-absorbing material is the single intended peak.
- Label mg vs. net peptide: like most research-market peptides, PT-141 is supplied as a lyophilized acetate salt, and counter-ion, residual water, and excipient weight all inflate gross vial mass. The net-peptide assay reports the actual milligrams of peptide so the labeled quantity can be checked rather than assumed.
What we measure on PT-141 (Bremelanotide)
Every PT-141 certificate reports exactly three results by reversed-phase HPLC with UV/diode-array detection at 214 nm. Identity is established by matching your sample's retention time and UV spectrum to a certified bremelanotide reference standard — a comparison that also distinguishes it from Melanotan II and from linear (uncyclized) analogs, which elute differently; chromatographic purity is reported as the area percent of the main peak relative to total integrated peak area; and the net-peptide assay reports the milligrams of peptide present per vial. The tryptophan in bremelanotide's sequence gives its UV spectrum useful character beyond the generic peptide-bond signal.

Can you really tell PT-141 apart from Melanotan II when they differ by so little?
Yes, with an honest caveat. The two differ by a C-terminal carboxylic acid (PT-141) versus amide (MT-II), and that single change alters polarity enough to shift reversed-phase retention time, so the two resolve against the certified bremelanotide standard under our method. The caveat: retention-time and UV-spectral matching is strong evidence but not a sequence proof — absolute confirmation of a one-group difference would take mass spectrometry, which is outside our scope.
Do you verify the cyclic lactam bridge?
Indirectly. An uncyclized (linear) analog is chemically different from the lactam-bridged molecule and does not co-elute with the certified cyclic standard, so a sample that matches the standard on retention time and UV spectrum is consistent with the intact cyclic structure. Direct structural proof of the bridge itself would require techniques beyond RP-HPLC-UV.
My vendor sells 'PT-141' and 'Melanotan II' from the same catalog — does that matter?
It is exactly the scenario where testing earns its keep. Compounds that sit next to each other in a catalog, share most of their structure, and are filled into identical-looking vials are the easiest to swap or mislabel, whether by error or intent. The identity result on your certificate tells you which compound your specific vial matches.
Will the net-peptide result match the 10 mg on my label?
The assay reports the milligrams of actual peptide measured in the vial you send, excluding acetate counter-ion, water, and excipient mass. Even honestly filled vials typically show some gap between gross labeled weight and net peptide; the certificate gives you the measured figure so you can judge whether the gap is normal salt-and-water weight or a genuine underfill.
