What Melanotan II is

Melanotan II (MT-II, MT-2) is a synthetic cyclic analog of alpha-melanocyte-stimulating hormone. Unlike the linear 13-residue Melanotan I, MT-II is a much shorter molecule: a heptapeptide core closed into a ring by a lactam bridge between side chains, with a C-terminal amide. That compact cyclic structure makes it chemically distinct from both Melanotan I and from PT-141 (bremelanotide), which is closely related to MT-II — PT-141 is essentially the des-amide (free carboxylic acid) counterpart. Because MT-II sits at the intersection of two confusion axes (MT-I vs. MT-II, and MT-II vs. PT-141) and is one of the most widely counterfeited compounds in the grey-market peptide category, it is a compound where an independent identity check does real work. Its tryptophan residue gives it useful UV character for diode-array comparison.

Why get your Melanotan II independently tested

  • The MT-I / MT-II axis: vendors and resellers routinely conflate the two Melanotans, and the molecules are structurally very different — linear 13-mer versus lactam-bridged cyclic heptapeptide. Retention-time and UV-spectral matching against a certified Melanotan II reference standard confirms which one is actually in the vial.
  • The MT-II / PT-141 axis: PT-141 (bremelanotide) differs from MT-II only at the C-terminus (acid instead of amide), and market price differences create a substitution incentive in both directions. The structural difference shifts chromatographic behavior, so a certified-standard comparison can flag a swap that the label would never reveal.
  • Counterfeit and underfilled vials: MT-II is a high-volume, heavily counterfeited product, which makes short-filled or padded vials a live risk. The net-peptide assay reports actual milligrams of peptide per vial — excluding acetate counter-ion, water, and excipient mass — for direct comparison against the labeled 10 mg.
  • Cyclization and synthesis impurities: forming the lactam bridge is an extra synthetic step, and failed or wrong-ring cyclization products, deletion sequences, and epimers can accompany the main product. Chromatographic purity as area percent shows how much of the UV-absorbing material is genuinely the main peak.

Every Melanotan II certificate reports three results by reversed-phase HPLC with UV/diode-array detection. Identity is a retention-time and UV-spectral match against a certified Melanotan II reference standard, which is the direct check against both MT-I and PT-141 substitution. Chromatographic purity is the area percent of the main peak relative to total integrated peak area, and the net-peptide assay reports milligrams of peptide per vial for comparison against the label.

Every result is issued as a certificate of analysis you — or anyone you forward it to — can verify cryptographically. Reporting identity, purity, and milligram content is the point: a vial can be pure and still underfilled, so we report all three. Melanotan II testing — FAQ

Can you tell Melanotan II apart from PT-141?

The two are close relatives — PT-141 is effectively the free-acid (des-amide) counterpart of MT-II — but they are distinct molecules whose difference is reflected in reversed-phase retention behavior. We compare your sample against a certified Melanotan II reference standard; material that is actually PT-141 is expected to fail the retention-time match. As always with our method, identity is a chromatographic and UV-spectral comparison, not a mass-spectrometry confirmation.

Can you tell Melanotan II apart from Melanotan I?

Yes. MT-I is a linear 13-residue peptide and MT-II is a cyclic heptapeptide; they behave differently under RP-HPLC and have their own reference standards. If your sample does not co-elute and spectrally match the MT-II standard, the certificate reports the non-conforming identity result. If you suspect you may actually have MT-I, tell us when ordering.

MT-II is supposedly counterfeited a lot — what does your test actually catch?

Three failure modes: wrong compound (the identity comparison against the certified standard), dirty product (chromatographic purity as area percent of the main peak), and short fill (net-peptide milligrams per vial versus the label). What it does not cover is anything outside RP-HPLC-UV scope — sterility, endotoxin, heavy metals, or definitive mass confirmation.

Does a high purity percentage mean my vial has the labeled 10 mg?

No — purity and content are independent results. Area-percent purity says how clean the peptide that is present is; the net-peptide assay says how much of it there is. A vial can be 99% pure and still contain 6 mg. That is why the certificate reports both, alongside identity.
