What Cagrilintide is

Cagrilintide, originally designated AM833 (also NNC0174-0833), is a synthetic amylin analog peptide. Structurally it is a 37-residue chain based on a pramlintide-like backbone carrying several amino-acid substitutions, and — critically for analysis — it is acylated at the N-terminal lysine with a C20 fatty-diacid side chain joined through a gamma-glutamic acid linker. That lipidation is the main structural feature separating it from its unacylated parent peptide, so confirming its presence is central to confirming identity. Cagrilintide is a research-grade compound and is not an approved drug product, which means material circulating on the research market carries no regulated guarantee of what is actually in the vial.

  • Acylation integrity: the defining feature of cagrilintide is its C20 fatty-diacid acyl chain. A truncated, des-acyl, or plain pramlintide substitute is a chemically different molecule, and its retention behavior on reversed-phase HPLC differs from genuine acylated cagrilintide — so identity testing against a reference standard helps flag an unacylated or wrong-backbone substitute.
  • Underfilling and content padding: amylin-class analogs are expensive and in high demand, creating economic incentive to underfill vials or bulk them out with excipient such as mannitol or buffer salts. A net-peptide assay reports the milligrams of actual peptide present rather than total powder mass, so you learn whether a labeled amount reflects real peptide.
  • Blend ratio verification: cagrilintide is frequently co-formulated with semaglutide and sold informally as 'CagriSema.' In a two-component blend the relative amount of each peptide matters, and chromatographic separation lets each peak be examined individually instead of trusting a stated ratio.
  • Purity and degradation: reconstitution, heat, and time can generate related-substance and breakdown peaks, and the acyl side chain adds further sites where related structures can appear. A chromatographic purity figure (area %) quantifies how much of the UV-absorbing material is the intended peptide versus co-eluting impurities and degradants.

What we measure on Cagrilintide

Each cagrilintide sample is analyzed by reversed-phase HPLC with UV/diode-array detection at 214 nm and compared against a certified reference standard. We report three results per certificate: identity (retention-time plus UV-spectral match to the reference standard), chromatographic purity expressed as area percent of the main peak, and a net-peptide assay giving the milligrams of peptide per vial. For a CagriSema-type blend, the components are resolved and reported separately where the method and available reference standards allow.

Every result is issued as a certificate of analysis you — or anyone you forward it to — can verify cryptographically. Reporting identity, purity, and milligram content is the point: a vial can be pure and still underfilled, so we report all three. Our scope — stated plainly

Testing is RP-HPLC-UV against a certified cagrilintide reference standard; because cagrilintide is a less-common peptide, a qualified reference standard may need to be sourced before your run, which we will confirm with you first. This method does not include mass spectrometry, sterility, endotoxin, heavy-metal, or residual-solvent testing.

Cagrilintide testing — FAQ

Can you tell genuine cagrilintide apart from pramlintide or a non-acylated version?

We distinguish material by matching retention time and UV spectrum to a certified cagrilintide reference standard. Because the C20 acyl chain shifts reversed-phase retention substantially, a des-acyl or wrong-backbone substitute typically fails to co-elute with the standard and is flagged on identity. Note that RP-HPLC-UV is not mass spectrometry — it does not measure exact molecular mass — so if definitive mass confirmation is required, that would need LC-MS, which we do not offer.

Can you verify a CagriSema (cagrilintide + semaglutide) blend?

Yes, where the two peptides resolve chromatographically. We report each component's peak and can provide a net-peptide figure for each where suitable reference standards are available. Reference standards for both cagrilintide and semaglutide may need to be sourced first, which we will confirm before running the sample.

Do you sell cagrilintide?

No. KMD Analytical is an independent, third-party testing laboratory and does not sell peptides. You mail in your own sample, and we return a certificate reporting its identity, chromatographic purity, and net peptide content.
