What PNC-27 is

PNC-27 is a 32-amino-acid synthetic peptide that joins a short segment of the p53 protein to a penetratin-derived sequence — a design intended to carry the p53 fragment across membranes. Analytically it is a long, amphipathic peptide with a high proportion of hydrophobic and basic residues, which means it retains strongly on a reversed-phase column, sticks to surfaces, and accumulates the usual long-synthesis byproducts: deletion sequences, incomplete couplings, and oxidation-prone residues. It is an investigational compound with no regulatory approval; material on the research-chemical market has no manufacturing or identity guarantee.

Why get your PNC-27 independently tested

Long-sequence purity: 32 residues by solid-phase synthesis produces truncation and deletion species that elute near the parent. Chromatographic purity as area percent reports how much of the UV-absorbing material is the full-length peptide.
Amphipathic handling loss: a peptide built to interact with membranes also interacts with vial walls and pipette tips. The net-peptide assay reports what is actually present in milligrams, against a reference standard, rather than what a fill weight implied.
Substitution on a high-priced compound: PNC-27 is expensive per milligram, which creates the incentive to replace it with a cheaper peptide of similar size. Identity by retention-time and UV-spectral match against a certified PNC-27 standard settles that.

What we measure on PNC-27

Every PNC-27 certificate reports identity (retention-time and UV-spectral match against a certified PNC-27 reference standard), chromatographic purity as area percent, and net-peptide content in milligrams per vial, by reversed-phase HPLC with diode-array detection at 214 nm, with 280 nm confirmation where the sequence's aromatic residues make it useful.

Every result is issued as a certificate of analysis you — or anyone you forward it to — can verify cryptographically. Reporting identity, purity, and milligram content is the point: a vial can be pure and still underfilled, so we report all three. Our scope — stated plainly

Testing is RP-HPLC-UV against a certified reference standard; a standard may need to be sourced before analysis, which can affect turnaround. KMD Analytical is an independent laboratory. We do not perform mass spectrometry, sterility, endotoxin, heavy-metal, or residual-solvent testing, and we make no assessment of biological activity.

KMD is an independent RP-HPLC-UV lab. We do not run LC-MS / mass-spec, sterility, endotoxin, or heavy-metal testing. If your job needs a capability we don't have, we'll say so.

PNC-27 testing — FAQ

Does the penetratin portion complicate testing?

It makes the peptide sticky and strongly retained, which we account for in sample handling and method conditions. It does not change what is reported: identity against the reference standard, purity as area percent, and milligrams per vial.

Can you tell PNC-27 from a truncated version?

Truncations and deletion sequences separate from the full-length peptide under reversed-phase conditions and appear as additional peaks, which lower the reported purity. We do not identify which residues are missing — that would need mass spectrometry — but the certificate shows how much of the material is not the main peak.

Is PNC-27 in your standing panel?

It is on our accepted list. Reference standards for less common peptides are sourced as needed, which can add a few days to the first order; subsequent orders run on the standard already in hand.
