What Humanin is

Humanin is a 24-amino-acid peptide encoded within the mitochondrial genome, and it is sold on the research market in more than one form: the native sequence (HN) and, more often, an analog with a single serine-to-glycine substitution at position 14 (S14G, usually labelled HNG or Humanin G). Those are different molecules with different retention behaviour, and vendors do not always say which one is in the vial. A 24-mer synthesised by solid-phase chemistry also carries the usual risks of deletion and truncation byproducts. Humanin is not an approved drug anywhere; material on the grey market has no regulated manufacturing or identity guarantee, which is exactly what independent testing is for.

Why get your Humanin independently tested

  • Variant confusion: native Humanin and the S14G analog (HNG) are frequently sold under one name. Identity is matched against the reference standard for the variant your label declares, so a mismatch between label and vial is reported rather than assumed away.
  • Sequence-level impurities: a 24-residue synthesis accumulates deletion sequences and incomplete-coupling byproducts that co-travel with the main peak. Chromatographic purity as area percent quantifies how much of the UV-absorbing material is the target peak.
  • Short fill and counter-ion weight: Humanin is a mid-priced peptide sold in small milligram quantities, where trifluoroacetate and water weight can be a large share of gross mass. The net-peptide assay reports the milligrams of peptide actually present.
  • Label claim you can hold someone to: a verifiable certificate that names the variant, the purity, and the milligrams is the only evidence that survives a vendor changing their story.

What we measure on Humanin

Every Humanin certificate reports identity (retention-time and UV-spectral match against the certified reference standard for the declared variant), chromatographic purity as area percent, and net-peptide content in milligrams per vial, by reversed-phase HPLC with diode-array detection at 214 nm. Tell us on the order form whether the label says Humanin, HNG, or S14G so the right standard is run beside your sample.

Every result is issued as a certificate of analysis you — or anyone you forward it to — can verify cryptographically. Our scope — stated plainly

Testing is RP-HPLC-UV against a certified reference standard for the variant named on your label; a standard for the less common variant may need to be sourced before analysis, which can affect turnaround. KMD Analytical is an independent laboratory. We do not perform mass spectrometry, sterility, endotoxin, heavy-metal, or residual-solvent testing, and we do not sequence peptides.

Humanin testing — FAQ

My vial just says Humanin — which standard do you use?

Ask your supplier which variant it is, and put that on the order form. Most grey-market Humanin is the S14G analog (HNG). If the label genuinely does not say, tell us and we will run the standard we hold and state on the certificate which variant identity was judged against.

Can HPLC tell native Humanin from HNG?

Under our method the two elute at different retention times, so a sample that matches one standard and not the other is reported as such. What HPLC-UV cannot do is sequence an unknown — it confirms or fails a match against a named reference, and the certificate says which.

What does a low purity result on Humanin usually mean?

Additional peaks near the main peak, most often deletion sequences from synthesis. Purity is the area percent of the main peak versus everything the column resolved; the chromatogram on the certificate shows where the rest of the area sits.
